Flow-Based CL-SMIA for the Quantification of Protein Biomarkers from Nasal Secretions in Comparison with Sandwich ELISA

Protein biomarkers in nasal secretions can be used as a measure to differentiate between allergies, airway diseases and infections for non-invasive diagnostics. The point-of-care quantification of biomarker levels using flow-based microarray facilitates precise rapid diagnosis displays the potential targeted effective treatment. For first time, we developed chemiluminescence sandwich immunoassay (CL-SMIA) interferon-beta (IFN-β) on Microarray Chip Reader-Research (MCR-R). Polycarbonate foils are cost-effective surface immobilizing capture antibodies. By commercially available set anti-human IFN-β antibodies, CL-SMIA compared directly an enzyme-linked immunosorbent assay (ELISA) performed microtiter plates concerning bioanalytical performance economic issues. Pre-incubation sample with detection antibodies lower consumption this allows longer interaction time antibody biomarker. direct injection pre-incubated samples into chips eliminates adsorption proteins tubing well contamination valves MCR-R clinical samples. small flow cell low volume 50 μL. limit 4.53 pg mL−1 was slightly increased ELISA which were 1.60 mL−1. possibility perform multiplexed mode makes it promising secretions.
